Structural analysis of PCNAs from Sulfolobus tokodaii


PCNAs are well-known to form a ring-shaped structure, even if the identity of amino acids sequence between them are low. In addition, PCNAs from crenarchaeal species are reported to form a heterotrimer consisting of PCNA1, PCNA2 and PCNA3, while in eukaryotes or euryarchaea PCNAs form a homotrimer. Interestingly, unique oligomeric state of PCNA that consisting of PCNA2 and PCNA3 without PCNA1 was found in Sulfolobus tokodaii.

We performed X-ray crystallographic analysis, small-angle X-ray scattering(SAXS) and gelfiltration analysis of PCNA2-PCNA3 complex from Sulfolobus tokodaii. Our presented study revealed that PCNA2-PCNA3 complex forms an elliptic ring-like heterotetramer in nature.
